目的:探讨LETM1蛋白过表达在子宫颈癌预后评估中的临床病理学意义。方法:实验共选取164例子宫颈活检及手术组织蜡块标本,其中包括94例宫颈鳞癌组织、12例宫颈腺癌组织、15例宫颈上皮内瘤变I级(CINⅠ)、22例宫颈上皮内瘤变II~III级(CINⅡ~Ⅲ)以及21例正常宫颈上皮组织。应用免疫荧光染色在宫颈癌He La细胞系中检测LETM1蛋白的定位情况,免疫组化法检测LETM1蛋白在上述宫颈病变组织中的表达,并分析其蛋白检测在宫颈癌预后评估中的意义。结果:免疫荧光结果表明,LETM1蛋白定位在宫颈癌He La细胞质及细胞膜上。免疫组化结果显示,LETM1蛋白在宫颈鳞癌和宫颈腺癌组织中表达率分别高达60.6%和75.0%,明显高于正常宫颈上皮组织及CINⅠ~Ⅲ(P〈0.05)。此外,LETM1蛋白在低分化宫颈癌组织中的阳性表达率明显高于中分化和高分化宫颈癌组织,且其阳性表达率在晚期宫颈癌组织中亦明显高于早期宫颈癌组织(P〈0.05),但LETM1蛋白的表达与宫颈癌患者年龄、肿瘤大小及有无淋巴结转移均无关联。结论:LETM1蛋白参与了宫颈癌的演进,其过表达可能预示宫颈癌的不良预后。
Objective: To explore the clinicopathological significance of LETM1 protein overexpression in prognosis evaluation of cervical cancer. Methods: A total of 164 cervical specimens were obtained after cervical biopsy and surgery,including 94 cases of cervical squamous cell carcinoma,12 cases of cervical adenocarcinoma,15 cases of cervical intraepithelial neoplasm Ⅰ( CINⅠ),22 cases of CIN Ⅱ-Ⅲ,and 21 cases of normal cervical epithelium. Immunofluorescent staining was used to detect the location of LETM1 protein in cervical cancer He La cells,immunohistochemical staining was used to detect the expression of LETM1 protein in the above- mentioned cervical specimens,the significance of LETM1 protein detection in prognosis evaluation of cervical cancer was analyzed. Results: Immunofluorescent staining showed that LETM1 protein located in cytoplasm and cell membrane of cervical cancer He La cells. Immunohistochemical staining showed that the expression rates of LETM1 protein in cervical squamous cell carcinoma and cervical adenocarcinoma were as high as 60. 6% and 75. 0%,respectively,which were statistically significantly higher than those in normal cervical epithelium and CINⅠ- Ⅲ( P 〈0. 05). In addition,the positive expression rate of LETM1 protein in poorly- differentiated cervical cancer was significantly higher than that in well- differentiated and moderately- differentiated cervical cancer,the positive expression rate of LETM1 protein in advanced cervical cancer was significantly higher than that in early cervical cancer( P 〈0. 05),but LETM1 protein expression was not correlated with age of patients,tumor size,and lymph node metastasis or not. Conclusion: LETM1 protein participates in the progression of cervical cancer,and its overexpression may indicate poor prognosis of cervical cancer.
