目的:研究胶质瘤中蛋白激酶P13K、AKT2 mRNA表达与NDRG2 mRNA表达的相关性,寻找与NDRG2相互作用的上下游分子。方法:用实时定量PCR技术检测29例脑胶质瘤及相应瘤旁组织中P13K、AKT2和NDRG2 mRNA表达量。结果:29例脑胶质瘤NDRG2 mRNA表达明显低于瘤旁组织(P〈0.05),且不同病理分级间表达有显著性差异(P〈0.05),其中Ⅳ级NDRG2 mRNA表达量明显低于Ⅱ级和Ⅲ级胶质瘤(P=0.007,P=0.03);AKT2,P13K mRNA表达在胶质瘤及瘤旁组织之间无明显差异(P〉0.05)。AKT2,P13K mRNA表达与NDRG2 mRNA表达的相关性无统计学意义(P〉0.05)。结论:NDRG2可能与胶质瘤的发生发展及恶性演变有关,在mRNA表达水平,还不能认为P13K、AKT2与NDRG2之间有相关性。
Objective:To investigate the expression and correlation of PI3K- AKT2 signal transduction pathway with NDRG2 in human glioma,so as to find the upstream or downstream molecular interacting with NDRG2. Methods: Real time PCR was used to analyze the expression of PI3K,AKT2 and NDRG2 mRNA in 29 glioma tissues and adjacent normal tissues. Results: Compared with adjacent normal tissues,the expression level of NDRG2 mRNA in glioma tissues was reduced significantly (0.95 ± 0. 16 vs 1.42 ± 0. 23, P 〈 0. 05). And there was a trend for NDRG2 level to reduce with increasing pathologic grade (P 〈 0.05). However, the expressions of AKT2 and PI3K had no obviously difference between glioma and adjacent normal tissues ( P 〉 0.05 ). The analysis of correlation had no statistial significance among PI3K, AKT2 and NDRG2 ( P 〉 0. 05 ). Conelution: The aberrant expression of NDRG2 is possibly associated with the malignant transformation of glioma, however,PI3K and AKT2 possibly have no correlation with NDRG2 in mRNA level in glioma.
