目的探讨虾青素(ATX)通过激活ERK1/2通路诱导血红素加氧酶-1(HO-1)表达的机制。方法ATX单独处理体外培养的SH—SY5Y细胞或运用ERK1/2抑制剂PD98059预处理,提取细胞蛋白,Western印迹方法检测pERK1/2和ERK1/2以及HO-1蛋白表达水平。结果与正常对照组相比,ATX可以时间依赖性和剂量依赖性诱导HO-1的表达上调,ATX时间依赖性和剂量依赖性激活ERK1/2,后者激活介导ATX诱导HO-1的表达上调,PD98059预处理后可以减弱ATx引起的HO-1蛋白表达水平降低。结论虾青素通过激活ERK1/2通路诱导HO-1的表达。
Objective To investigate whether Astaxanthin (ATX) induce Heme oxygenase-I expression through activating ERK1/2 pathway. Methods Cultured SH-SY5Y cells were treated with ATX preincubated with or without ERK1/2 inhibitor,PD98059. The levels of pERK1/2 ,ERK1/2, and HO-1 proteins were measured by Western blot. Results ATX induced the expression of HO-1 in a concentration- dependent and increased the expression of HO-1 protein in time-dependent. ATX activated the ERK MAPK cascade. ATX-induced HO-1 expression was blocked by ERK1/2 inhibitor PD98059 in concentration-dependent manner. Conclusions ATX induces Heme oxygenase-1 expression through activating ERKI/2 pathway
