为建立中药活性成分的抗体测定技术平台,将中药活性成分芍药苷与人血清白蛋白偶联成完全抗原(半抗原-蛋白质复合物),并利用基质辅助激光解吸电离-飞行时间-质谱(MALDI-TOF-MS)技术测定了上述人工抗原的基本质谱特征,求得了芍药苷-人血清白蛋白复合物的半抗原数(hapten number)为12.试验结果表明,利用MALDI-TOF-MS方法测定人工合成的半抗原-蛋白质复合物的质谱特征等,具有所需样品量少、结果准确和迅速可靠等优点,为药用植物活性成分的抗体技术研究提供了科学方法和试验依据,对于判断人工抗原的质量具有科学意义.
In the establishment of monoclonal antibody technology platform, hapten - carrier protein conjugates of a bioactive constituent paeoniflorin (PF) from traditional Chinese herbal medicine and human serum albumin (HSA) were artificially synthesized, and the hapten number of the PF-carrier protein conjugates (PF-HSA) was determined successfully with MALDI-TOF-MS. The results showed the hapten number of the synthesized PF-HSA was 12. This approach would help to determine the quality of artificial antigen, and contributes to antibody preparation of anti - bioactive constituents from traditional Chinese medicines.
