利用杂交瘤技术制备了以甘草中主要活性成分甘草酸为半抗原的单克隆抗体,建立了以抗甘草酸单克隆抗体为基础的ELISA分析方法,测定了10种中成药的甘草酸含量.结果表明,不同产家与剂型的中成药的甘草酸含量差异极大,约在28—1900μg/mL范围.与HPLC测定结果比对结果显示,两者的测定结果相近,大多在±5%范围之内,证明ELISA技术对中成药主要活性成分的分析测定是可行的,对HPLC测定提供了有益的技术补充和验证手段.
A monoclonal antibody (MAb) against the bio-active constituent glycyrrhizic acid (GA) of Glycyrriza uralensis was prepared using hybridoma techniques, and an enzyme-linked inmmnosorbent assay (ELISA) based on the anti-GA MAb was also established to determine the GA content in ten traditional Chinese prepared medicines. The results showed that the GA contents ranged from 28 to 1 900 μg/ mL in these medicines of different manufacturers and pharmaceutical preparations. Comparison of the error rates of GA determination between ELISA and HPLC demonstrated high similarity, mostly less than ± 5%. The results verified the feasibility of immunoassay ELISA in the determination of major bio-active constituents from the traditional Chinese prepared medicines.
