目的:研究芪玄抑甲宁对甲亢大鼠Th17细胞相关因子的调控影响。方法:将雌性SD大鼠随机分成空白组(生理盐水组)和模型组,以尾静脉注射小肠结肠炎耶尔森氏菌造模,模型组大鼠随机分为模型组、芪玄抑甲宁14.4 g/kg、7.2 g/kg、3.6 g/kg剂量组和甲巯咪唑组(3mg/kg)。用酶联免疫法检测大鼠血清中IL-17、IL-6、TGF-β水平,免疫组化法和Real-time PCR法检测甲状腺组织中IL-17、IL-17R和IL-17mRNA、IL-17RmRNA的蛋白表达。结果:芪玄抑甲宁(14.4 g/kg)可以明显降低甲亢大鼠血清中IL-17、IL-6和TGF-β的水平,回调甲亢大鼠甲状腺组织中IL-17、IL-17R和IL-17mRNA、IL-17RmRNA的蛋白表达水平。结论:芪玄抑甲宁对Th17细胞的相关因子具有调控影响,这可能是芪玄抑甲宁治疗甲亢的重要作用机制之一。
Objectives:To study the regulatory effect of Qixuanyijianing on Th17 cells of rats with hyperthyroidism.Methods:60 SD female rats were divided into control group and hyperthyroidism groups.Hyperthyroidism rats were copied by injecting Yersinia enterocolitica into rats’caudal vein.The hyperthyroidism rats were randomly divided into model group,14.4 g/kg,7.2 g/kg,3.6 g/kg dose group of Qixuanyijian-ing and positive group(3mg/kg).The levels of IL-17,IL-6 and TGF-βwere measured by ELISA.Immunohistochemistry method and the Real-time PCR were used to measure the level of IL-17,IL-17R and IL-17mRNA,IL-17RmRNA protein expression of thyroid tissues.Results:Qixuanyijianing-group(14.4 g/kg)can reduce the serum level of IL-17,IL-6,TGF-βof the hyperthyroidism rats and IL-17 in thyroid tissue,IL-17 R and IL-17 mRNA,significantly decrease the protein expression level of IL-17RmRNA,which show the regulatory effect of Qixuanyijianing to Th17 cells.Conclusions:Qixuanyijianing has an regulatory influence on Th17 cells,which may be an important mechanism of Qixuanyijianing on hyperthyroidism.
