高甘油三酯血症中对氧磷酯酶-1活性的研究

目的:观察高甘油三酯血症(HTG)状态下高密度脂蛋白(HDL)相关抗氧化酶——对氧磷酯酶-1(PON-1)的活性变化。方法:收集湖北武汉地区40例单纯性HTG患者和65例正常人群的血样,比较血脂分布特征,利用酶水解底物对氧磷的原理测定血浆HDL特征性PON-1的活性;同时以脂蛋白脂肪酶基因敲除(LPL-/-)鼠为HTG动物模型,检测血清中甘油三酯(TG)、总胆固醇(TC)含量和PON-1的活性改变。结果:①HTG组与对照组相比,血浆载脂蛋白(apo)B100水平显著升高(P〈0.01),HDL-C、apoA-Ⅰ水平降低(P〈0.05);PON-1酶活性显著降低(P〈0.001);②HTG合并正常HDL-C组与低HDL-C对照组比较,PON-1活性无明显差别,但PON-1/apoA-Ⅰ比值显著降低(P〈0.05);③LPL-/-鼠与对照组比较TG明显升高(P〈0.001),PON-1活性显著下降(P〈0.001)。结论:血清PON-1活性降低反映HDL抗氧化功能的异常,这可能与HTG易致动脉粥样硬化的发生密切相关。

英文摘要:

Objective:To investigate the alteration of the activity of paraoxonase-1(PON-1),an apoli-poprotein AⅠ(apoAⅠ)-associated arylesterase which located on the high density lipoprotein (HDL),in hypertriglyceridemia(HTG).Methods:The fasting plasma samples were obtained from 40patients with HTG patients and 65 control individuals of Wuhan,Hubei province,the plasma lipid profile including triglycerides (TG),total cholesterol(TC),HDL cholesterol (HDL-C),low-density lipoprotein cholesterol(LDL-C),apoA-Ⅰand apoB100were determined enzymatically,and plasma PON-1activity was measured by monitoring the increase of absorbency at 405nm using paraoxon as the substrate.Lipoprotein lipase gene knockout(LPL-/-)mice as HTG models were also detected.Results:Compared with control subjects,the level of plasma HDL-C and apoA-Ⅰof HTG group were lower(P0.05),PON-1activity was also lower obvi-ously(P0.001),and the level of apoB100was higher(P0.01);plasma PON-1activity was not significantly different between lower HDL-C with normal TG group and HTG with normal HDL-C group,but PON-1/apoA-Ⅰ ratio significantly decreased in HTG group(P0.05);com-pared with that respectively in wild type mice,significantly higher TG and lower PON-1 activity were detected in LPL-/-mice(P0.001).Conclusion:The decrease of plasma PON-1activity that reflects abnormality of HDL anti-oxidation might be closely relative to atherogenesis in HTG patients.
