目的观察当归补血汤对骨髓辐射损伤过程中核因子(NF)-κB及下游因子bax、bcl-2表达的影响。方法取原代培养7 d的骨髓基质细胞用137Cs-γ射线(4 Gy)照射,在照射后24 h用含不同剂量的当归补血汤载药血清干预24 h;小鼠同剂量γ射线照射后,灌胃给药同剂量的当归补血汤1 w,处死小鼠取骨髓。应用免疫荧光技术,real-time RT-PCR分别检测辐射损伤骨髓基质细胞模型和小鼠模型骨髓的NF-κB及下游因子bax、bcl-2的表达。结果辐射可激活NF-κB信号,启动MSC和骨髓细胞凋亡过程。经当归补血汤干预,MSC和骨髓细胞内可下调NF-κB基因的表达,但随着药物剂量加大下调趋势减缓(P<0.05);当MSC和骨髓细胞内bcl-2/bax比值升高时,对促凋亡信号敏感性减弱,表现出抗凋亡的特点,细胞存活寿命延长(P<0.05)。结论当归补血汤可通过影响NF-κB的表达调节bcl-2/bax,减少骨髓内细胞凋亡,有利于辐射损伤后机体造血免疫机能的恢复。
Objective To investigate the effect of Danggui buxue decoction on inhibiting bone marrow from apoptosis.Methods Bone marrow stromal cells ( BMSCs) from mice were primary cultured for 7 days and then radiated by 137Cs-gamma ray (4Gy) .After irra-diation for 24 h, BMSCs were cultured in the different concentration serum containing drug of Danggui buxue decoction for 24 h.After radia-ted by the same dose of gamma ray, mice were given the same dose of Danggui buxue decoction by intragastric administration for one week , and then sacrificed and extracted bone marrow.NFk-appa B and its downstream molecule bax and bcl -2 expressions of BMSCs and mice bone marrow radiated by gamma ray were detected separately by immunofluorescence technique and real -time fluorescence quantitative RTP-CR method.Results NF-kappa B signal activated by radiation could start the process of BMSCs and bone marrow cells apoptosis .BMSCs and bone marrow cells down regulated the expression of NF-kappa B after the intervention of Danggui buxue decoction;but with dose increasing, a downward trend was slowed (P<0.05).When the bcl-2/bax ratio increasing in BMSCs and bone marrow cells, the apoptotic signals sensi-tivity was decreased and cells showed the anti-apoptotic specialty and life extension(P<0.05).Conclusions Danggui buxue decoction could affect the expression of NF-kappa B and then adjust the bcl-2/bax ratio in order to reduce cell apoptosis in bone marrow radiated by gamma ray, and furthermore is conducive to the recovery of hemopoiesis and immune function after radiation injury.
