目的:对阿魏属4种药用植物基因组总DNA提取方法进行比较,建立阿魏属药用植物总DNA的最佳提取方法,为下游分子生物学实验提供依据。方法:利用3种不同的DNA提取方法提取阿魏属4种药用植物的总DNA;用核酸蛋白检测仪和琼脂糖凝胶电泳方法检测所得总DNA的浓度和纯度;用ISSR分子标记的方法检测所得总DNA的质量。结果:试剂盒法提取的总DNA纯度最高,质量最好,能够将所有样品扩增出丰富而清晰的条带。所需时间短,操作简单。改良的CTAB法与SDS法提取的总DNA纯度及质量均次于试剂盒法,所需时间长,操作复杂。结论:试剂盒法为提取阿魏属4种药用植物总DNA的最佳方法,能够直接用于下游分子生物学实验。
Objective: For providing the basis for downstream molecular biology experiments, to determine the optima extraction method of genomic DNA from four traditional medicine, which belong to the genus Ferula by comparing the different extraction methods, genomic DNA Kit method, the improved CTAB method and improved SDS method. Methods: Genomic DNA was extracted by three different methods and the purity and concentration of DNA were compared by UV and gel electrophoresis. The qualities of DNA were analyzed by ISSR. Results: Genomic DNA Kit method was higher than the improved CTAB method and improved SDS method with more quality, more clear bands, shorter time and more simple operation. Conclusion: Genomic DNA Kit method is the optimal method for plant medicines which belonged to the genus ferula and can be used directly in downstream molecular biology experiments.
