目的从离体细胞研究层面,研究中药密蒙花方对高糖状态下视网膜Mfiller细胞损伤的保护作用。方法(1)采用大鼠视网膜Mfiller细胞在30mmol/L高糖下进行体外培养,模拟高糖环境,再分别给予20%含密蒙花方血清(以下均称合药血清)干预12h,24h、36h,观察密蒙花方含药血清对大鼠视网膜Mtiller细胞增殖的影响。(2)采用流式细胞仪AnnexinV—FITC/PI双色染色法观察密蒙花方对高糖状态下视网膜Mtiller细胞凋亡的影响。结果(1)高糖组较正常组吸光度(OD)值增高;含药血清组与高糖组相比,0D值明显降低,各合药血清组间比较,随着时间的延长,OD值逐渐降低,差异具有统计学意义(P〈0.05)。(2)高糖组较正常组细胞早期凋亡比例减少,活细胞比例增高,各含药血清组与高糖组相比,细胞早期凋亡比例增高,活细胞比例减少(P〈0.05)。密蒙花方通过诱导早期高糖状态下视网膜Mtiller细胞的早期凋亡,抑制细胞反应性增生,从而起到防护高糖状态下视网膜神经细胞损伤的作用。结论密蒙花方可以抑制早期高糖状态下视网膜Mfiller细胞的增殖。
OBJECTIVE To investigate the effect of Mimeng flower decoction on the retina Mtiller cell damaged by high glucose. METHODS Rat retina Mtiller cells were cultured with 30 mmol/L glucose, and were treated with serum medium containing 20% Mimeng flower decoction for 12, 24 or 36 h. Detected the effect of Mimeng flow- er decoction on the proliferation of the rat retina Miiller cells cultured with high glucose by the way of CCK-8. De- tected the effect of Mimeng flower decoction on the apoptosis of the rat retina Mtiller cells cultured with high glucose by flow cytometry Annexin V-FITC/PI two-color staining. RESULTS (1) The absorbance value of high glucose group was higher than normal group(P〈0.05 ). Absorption value of the rat retina Mtiller cells treated by serum medium containing Mimeng flower decoction was declined compared with model group (P〈0.05). (2)Compared with normal group, the cell early apoptosis proportion of high glucose group was reduced, live cell ratio was increased and the difference of changes was significant (P〈0.01). Compared with high glucose group, the cell early apoptosis proportion of each Mimeng flower group was increased, live cell ratio was decreased (P〈0.05). CONCLUSIONS Mimeng flower decoction could inhibit retina Mtiller cell proliferation under high glucose. Mimeng flower decoction could inhibit cell hyperplasia through promote early apoptosis of the retina Mtiller cell under high glucose.
