肿瘤严重危害人类健康,早期诊断、早期治疗可显著降低患者的死亡率,是肿瘤防治研究方向的重点,而深入认识癌症的发病原因和机制则是实现癌症早期诊治的关键。近年来,新兴的蛋白质组学技术可从整体上研究疾病的发生机制,寻找与疾病相关的标志蛋白,为肿瘤研究提供了新思路和新平台。蛋白质组学可以大规模地定量分析细胞内蛋白质的表达水平变化、翻译后修饰变化等,并可深入研究蛋白质间的相互作用,非常有利于发现调控肿瘤进程的关键因子。本文概述了目前蛋白质组学研究中的主要策略和技术,及其在肿瘤标记物筛选和作用机制研究中的应用进展。
Tumor has serious impact on people's health.Early diagnosis and early treatment can significantly reduce the mortality of patients, which is the key of tumor prevention.In-depth understanding of tumor developing mechanism is the key point of tumor diagnosis and treatment.In recent years, the emerged proteomics technology could profile the dynamic changes of global proteins in cells and screen markers associated with disease, which provides a new insight and technology platform for tumor research.Protemics could in large scale quantitation the change of intracellular protein expression level, the modification after translation, and in-depth understanding of the interaction between proteins, which is very helpful in finding key molecules in tumor development.This article mainly focus on the current strategy in proteomics research, as well as its application progress in screening of tumor markers and exploring of tumor mechanism research.
