采用新设计COI、16S、CR3种线粒体基因(位点)的简并引物,对江西地区常见蛇类(2科6属7种)共20个个体样本进行了DNA条形码分析的初步尝试,将实验结果结合GenBank部分蛇类序列数据分析表明,在本实验涉及的蛇类个体及类群的识别上,16S rRNA基因片段合乎通用条码标记的序列的要求;且本实验设计的16S简并引物有适用性强、宽容度大的优势。其次,本研究结果支持"16S rRNA基因至少应该作为脊椎动物标准条码标记———线粒体COI基因不可缺少的补充"观点;提示当前对DNA条形码在不同生物类群的应用方面,尚待在更大的标本样品数量及更多不同基因片段序列数据的支撑。
Referred to snake sequence data from GenBank,and studied 20 samples of snakes in Nanchang,which include 2 families,6 genus,and 7 species,to present a first attempt in applying the usage of DNA barcode in the species of snakes.Our experiment demonstrates,in terms of identifying snakes samples or groups,mitochondrial 16S rRNA gene fulfills the requirements for a universal DNA barcoding marker;we also show certain advantage in the application and universality of combination.Furthermore,our result supports the opinion:"We strongly advocate the use of 16S rRNA as standard DNA barcoding marker for vertebrates to complement COI,especially if samples a priori could belong to various phylogenetically distant taxa and false negatives would constitute a major problem."(Miguel et a1.2005).we recommend richer species samples and larger various gene sequences to support a wider application of DNA barcode in different biological field.
