目的:探讨量子点标记大鼠肌源性干细胞的可行性。方法:采用胶原酶、dispase和胰酶消化分离新生SD大鼠腓肠肌,差速贴壁法获取PP6细胞,倒置显微镜下观察细胞的形态并应用免疫组化法对其鉴定;应用ZnS包被的CdSe量子点对固定后细胞进行标记,荧光显微镜下观察。结果:通过差速贴壁法获取的PP6细胞经过免疫方法鉴定,证实为肌源性干细胞;量子点标记后的细胞在荧光显微镜下呈现橘红色荧光。结论:量子点标记肌源性干细胞具有可行性,为今后活体示踪研究打下了基础。
Objective:To investigate the feasibility of labeling rat muscle-derived stem cells with quantum dots(QDs).Methods:After the digestion with collagenase,dispase and trypsin,neonatal SD rat's gastrocnemius was separated into single muscle cells,and then the cells were purified by apreplate technique.The cells were observed with inverted microscope,and identified with immunohistochemistry.Then the fixed cells were labeled with CdSe QDs.Results:The preplated PP6cells were muscle derived stem cells.The labeled stem cells showed orange-red fluorescence in the fluorescence microscope.Conclusion:It is feasible that muscle-derived stem cells are marked with QDs,and this study paved the way for our future in vivo tracer studies.
