目的探讨大鼠肌源干细胞(MDSCs)的分离、培养新方法,并应用量子点进行标记。方法应用差速贴壁法对10只新生SD大鼠腓肠肌进行原代培养获取肌源干细胞(MDSC),然后量子点对其进行标记,荧光显微镜下观察效果。结果应用新生SD大鼠取材,消化时间25min,可明显提高获取细胞数量(每高倍视野增加约25%);荧光显微镜下观察量子点标记后的肌源干细胞,紫外光激发(激发波长≤400nm)呈现明亮的橘红色。结论应用新生SD大鼠取材缩短消化时间可明显提高肌源干细胞的获取量,量子点可成功标记肌源干细胞。
Objective To investigate new methods for isolation and culture of rat muscle-derived stem cells ( MDSCs), and then label them with quantum dots. Methods In 10 neonatal SD rat skeletal muscle, MDSCs were purified by using the preplate technique. After labeling them with quantum dots, we observed the result under the fluorescence microscopy. Results In the neonatal SD rats, the number of MDSCs was increased by about 25% in a high-powered field under the digestion time of 25 rain. Under the fluorescence microscopy, quantum dot-labeled MDSCs displayed bright orange-red fluorescence. Conclusion By using neonatal SD rats and 25min digestion time, MDSCs can be obtained largely and quantum dots can successfully mark MDSCs.
