目的探讨血管内皮生长因子(vascular endothelial growth factor,VEGF)基因多态性与抑郁症的关联及其与环境交互作用在抑郁症发病中的作用。方法提取274名汉族单相抑郁症患者(病例组)和273名汉族健康人(对照组)的全基因组DNA,应用单碱基延伸SNP分型技术(SNaP—shot)方法检测病例组和对照组VEGF.2578G/A位点的基因型和等位基因频率,应用二项Logsitic回归模型进一步分析VEGF-2578C/A基因和负性生活事件的交互作用与抑郁症的关联。结果病例组和对照组VEGF-2578C/A基因型A/A、A/C、C/C频率分布为4.0%、37.6%、58.4%和6.6%、35.5%、57.9%;等位基因A、C频率分布为22.8%、77.2%和24.4%、75.6%,两组间均差异无统计学意义(χ^2=1.880,P=0.391;χ^2=0.364,P=0.546)。VEGF-2578C/A多态性和负性生活事件交互作用与单相抑郁症的关联无统计学意义(P=0.716)。结论VEGF-2578C/A基因多态性及其与环境因素交互作用与抑郁症不存在关联。
Objective To investigate the correlation between VEGF-2578C/A gene polymorphism and depression, and to reveal the mechanism of the interaction between enviroment and VEGF gene polymorphism in depression patients. Methods The genomic DNA were extracted from 274 patients with depression (case group) and 273 healthy subjects (control group).Snapshot technique was used to detect the genotype and allele frequency of VEGF-2578C/A gene. The logistic regression model was used to analyze the interaction between VEGF-2578C/A gene polymorphism and negative life events. Results The frequency of VEGF-2578C/A genotype A/A, A/C and C/C in the case group were 4.0%, 37.6%, 58.4% respectively while in the control group were 6.6%, 35.5%, 57.9% respectively.The allele frequency of A was 22.8% in ease group and 24.4% in control group while the allele frequency of C was 77.2% in case group and 75.6% in control group.There was no significant difference in the frequency of VEGF-2578C/A genotype and allele frequency between the case group and control group (χ^2= 1.880, P=0.391 ;χ^2 =0.364, P=0.546). The interactions between VEGF-2578C/A gene polymorphism and negative life events were not statistically significant ( P= 0.716). Conclusion The interaction between VEGF-2578C/A gene and environmental factors may not be associated with depression.
