目的 探讨五羟色胺2A受体(5-hydroxytrytamine receptor 2A,5-HTR2A)基因多态性、负性生活事件交互作用与抑郁伴自杀的关系.方法 提取中国北方地区130名抑郁伴自杀者(病例组)和118名抑郁无自杀者(对照组)基因组DNA,采用Snapshot技术分别扩增rs6311和rs6313位点的基因组DNA片段,将PCR产物直接测序检测病例组和对照组的基因型,应用二分类Logistic回归模型分析rs6311、rs6313与负性生活事件的交互作用.结果 病例组与对照组rs6311位点的基因型(x2=0.012)和等位基因频率(x2 =0.062)比较均差异无统计学意义(P>0.05).两组rs6313位点的基因型比较,差异无统计学意义(x2=1.035,P>0.05),但等位基因频率比较差异有统计学意义(等位基因A:73.1% vs 50.4%;等位基因G:26.9%vs 49.6%;x2=27.029,P<0.05).rs6311和负性生活事件交互作用差异有统计学意义(OR=6.155,95% CI=1.372~27.612,P<0.05).rs6313和负性生活事件交互作用差异无统计学意义(OR=0.772,95% CI=0.231~2.579,P>0.05).结论 rs6311与负性生活事件的交互作用在抑郁伴自杀的发病机制中具有重要意义.
Objective To explore the relationship between the interaction of 5-hydroxytrytamine receptor 2A (5-HTR2A) gene polymorphism and negative life events and depression with suicidal tendency.Methods Extraction genomic DNA from 130 patients of depression with suicidal (case group) and 118 patients of depression with non-suicidal(control group) in northern China.Snapshot technology were used to amplify genomic DNA fragments from rs6311 and rs6313 sites,the PCR products were sequenced directly to detect genotype case and control groups,and logistic regression model was used to analyze the interaction between rs6311,rs6313 and negative life events.Results The rs6311 locus genotype (x2 =0.012) and allele frequencies (x2 =0.062) were not statistically significant between case group and control group (P〉0.05).The rs6313 locus genotype (x2=1.035) showed no significant difference(P〉0.05),but the allele frequencies were statistically significant (allele A:73.1% vs 50.4%;allele G:26.9% vs 49.6%) (x2=27.029,P〈0.05).The interaction of rs6311 and negative life events were statistically significant (OR=6.15,95% CI =1.372-27.612,P〈0.05).The interaction of rs6313 and negative life events were not statistically significant (OR=0.772,95% CI =0.231-2.579,P〉0.05).Conclusion The interaction of rs6311,rs6313 and negative life events plays important roles in patients of depression with suicidal.
