目的研究肝癌血管生成拟态(VM)形成与肝癌干细胞的关系。方法在体外分别建立肝癌细胞株HCC97H、SMMC7721以及正常肝细胞L02三维培养,观察它们各自形成VM的能力。利用激光捕获显微切割技术分离出形成VM的肝癌细胞,RT-PCR和Western blot分别检测内皮相关分子VE-cadherin、CD31及肝癌干细胞标志物CD133和CD34表达水平。结果①HCC97H细胞在三维培养下形成VM;SMMC7721和正常肝细胞L02不形成VM;②激光捕获显微切割成功捕获目的细胞;③各组中VE-cadherin mRNA和蛋白的表达差异有统计学意义(P〈0.05)。CD31在肝癌细胞中不表达。CD133 mRNA和CD34 mRNA在各组中表达差异有统计学意义(P〈0.05);CD133和CD34蛋白在各组间表达差异有统计学意义(P〈0.05)。结论肝癌干细胞表达与肝癌细胞VM的形成有关。
Objective To study the relationship between liver cancer stem cells and vasculogenic mimicry(VM).Methods Three-dimensional cell cultures were established in several cell lines of HCC97H,SMMC7721 and L02 cell,and observed the ability to form VM.Interesting cells which formed VM were isolated by the laser capture microdissection,and endothelial related molecules of VE-cadherin,CD31 and liver cancer stem cells markers CD133 and CD34 were analyzed by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and Western blot subsequently.Results HCC97H cell had the ability to form VM in three-dimensional cell cultures,while SMMC7721 and L02 cell hadn’t the ability.Interesting cells had been captured by LCM successfully.In comparative analysis,the expression level of VE-cadherin gene and protein in different groups were statistically significant(P 〈0.05),whereas CD31 was not expressed in the liver cancer cell lines.The mRNA and protein expressions of CD133 and CD34 in different groups were statistically significant(P 〈0.05).Conclusion Liver cancer stem cells may have impact on the formation of VM in hepatocellular carcinoma.
