siRNA作为基因功能研究和临床治疗的重要工具,已经引起了科学界的广泛关注,世界上许多学术和商业机构也开发了相应的siRNA设计程序.近年来,随着对siRNA机制研究的不断深入,尤其是2004年Reynolds提出siRNA理性设计规则之后,许多新的理论极大地推动了siRNA设计程序的发展.本文根据2003至2006年以来对siRNA机制的研究进展,在原有的设计程序siRNApro的基础上进行了更新并推出了2.0版,为siRNA作用机制的研究和siRNA相关产品的开发提供更优质的理性设计服务.
As an important tool of gene function studies and clinical therapy, siRNA has drawn extensive attentions in the biomedical research fields. After the discovery of RNA interference phenomenon in 1998, numerous researchers have closely investigated the mechanism of siRNA and attempted to formulate the rules for siRNA designs which have already been adopted in many siRNA designing programs. The outcome of siRNA design has been dramatically improved by the rational siRNA design rules summarized by Reynolds, et al. since 2004. The Bioinformatics Centre of South China University of Technology has built a siRNA designing system "siRNA pro" in 2003. Currently, to provide better services for the siRNA-related researches, an update of this program which is based on the latest developments of the designing rules is urgently demanded. siRNA pro 2.0, which has combined and taken advantages of the basic empirical rules, the rational design rules, the target mRNA accessibility rules the siRNA duplex thermodynamics rules, and the siRNA specificity rules, will ultimately help investigators to obtain highly effective and specific siRNA designs.
