Cyclopamine调控Hedgehog信号通路对正常角质形成细胞的影响

中文摘要:

探讨cyclopamine抑制Hedgehog信号转导通路对角质形成细胞增殖的影响。方法:在培养的Hacat角质形成细胞中加入Hedgehog信号转导通路特异性抑制剂cyclopamine,在不同的作用时间,应用MTT和BrdU掺入的方法,检测cyclopamine对Hacat角质形成细胞生长和增殖的影响。结果:cyclopamine可以抑制Hacat角质形成细胞的生长和增殖。结论:抑制Hedgehog信号转导通路可能对角质形成细胞增殖起到一定的抑制作用。

英文摘要:

Objective To investigate effect of cyclopamine on proliferation of Hacat cells (keratinocyte) by regulating Hedgehog signaling pathway. Methods The Hacat cells was incubated with cyclopamine, which is the specific inhibitor of Hedgehog signaling pathway, The Hacat cells" proliferation was measured with MTT and BrdU assay at different time after the cells were incubated with cyclopamine. Results Cyclopamine could inhibit the proliferation of Hacat cells compared with the control groups. Conclusion Cyclopamine can inhibit the proliferation of keratinocyte by inhibiting Hedgehog signaling pathway.
