目的:探讨Hedgehog信号通路特异性抑制剂cyclopamine对人正常角质形成细胞HaCaT细胞白介素(IL)-8分泌的影响。方法:在HaCaT细胞培养基中加入cyclopamine,采用反转录(RT)-PCR和ELISA方法检测HaCaT细胞在cyclopamine处理后不同时段IL-8 mRNA及其分泌的变化。结果:HaCaT细胞在cyclopamine处理后IL-8的mRNA表达及其分泌均减少(P〈0.05)。结论:Hedgehog信号通路特异性抑制剂cyclopamine可抑制HaCaT细胞分泌IL-8。
Objective: To investigate the influence of cyclopamine, a specific inhibitor of Hedgehog pathway, on the secretion of interleukin 8 (IL-8) in HaCaT cells. Methods: RT-PCR and ELISA were employed to measure the secretion of IL-8 in HaCaT cells at different time after treatment with cyclopamine. Results: IL-8 mRNA expression was significantly inhibited in HaCaT cells treated with cyclopamine, and IL-8 secretion was also reduced (P 〈 0.05). Conclusion: Cyclopamine, a specific inhibitor of Hedgehog pathway, can suppress the IL-8 secretion of HaCaT keratinocytes.
