目的探讨缺氧诱导因子-1α(HIF-1α)与上皮型钙黏素(E-cadherin)在肝细胞癌中的表达、相互关系及其临床意义。方法免疫组化法检测100例临床肝癌标本及10例正常肝组织标本中的HIF-1α和E-cadherin的表达,分析其表达与肝癌临床病理特征的相关性;用Western blot方法检测不同转移潜能肝癌细胞株HIF-1α和E-cadherin的表达,分析其表达与肝癌转移潜能的相关性。结果免疫组化检测发现HIF-1α在肝癌中存在高表达(80/100,80%),与肿瘤分期、侵袭转移密切相关(P=0.002,P=0.018);52例肝癌组织中存在E-cadherin表达降低,与肿瘤侵袭转移亦存在明显相关性(P=0.001);在肝癌组织中HIF-1α与E-cadher-in表达存在着显著负相关(P=0.000)。Western blot检测提示高转移潜能肝癌细胞HIF-1α表达较低转移肝癌细胞明显升高而E-cadherin表达却明显降低。结论 HIF-1α、E-cad-herin异常表达与肝癌进展及转移密切相关,HIF-1α及E-cadherin有望作为肝癌恶性潜能判断的重要参考指标。
Objective To investigate the expression of hypoxia-inducible factor 1 alpha(HIF-1α) and E-cadherin,analyze their correlations and clinical significance in hepatocellular carcinoma(HCC) cases.Methods The expression profiles of HIF-1α and E-cadherin were detected on 100 cases HCC clinical samples and 10 normal liver tissues by immunohistochemical staining method,and their correlations with clinicopathological parameters were analyzed.The expression of HIF-1α and E-cadherin in HCC cells with different metastatic potentials was examined by Western blot method.Results HIF-1α expression was positive in 80% HCC patients,which was associated with TNM stage,invasion and metastasis(P=0.002;P=0.018);however,52% HCC showed E-cadherin expression reduced,its expression was also associated closely with invasion and metastasis(P=0.001).In HCC,HIF-1α expression was negatively correlated with E-cadherin expression(P=0.000).Western blot analysis showed that the HIF-1α expression in highly metastatic HCC cells was significantly higher than that in lowly metastatic HCC cells.Whereas E-cadherin expression was lower in highly metastatic HCC cells.Conclusion Abnormal HIF-1α and E-cadherin expressions are associated with HCC progression and metastasis;HIF-1α and E-cadherin expression can serve as important markers for judgment of HCC metastasis potency.
