为了建立紫苏草离体试管开花体系,本试验以紫苏草种子无菌苗为外植体,通过对培养基中激素及营养水平的调整、采用低温处理和PEG的使用,从中找到离体开花的最适条件。结果表明:植物激素影响紫苏草试管花形成发育,采用MS附加6-BA 1.0 mg·L^-1和IAA0.5 mg·L^-1对于紫苏草试管苗开花的诱导效率最高;采用10℃低温处理2 d对试管苗开花率有提高作用;培养基中营养水平不同对开花有影响,半量MS培养基利于试管开花;在培养基中加入10%PEG-6000可以提高开花率和开花质量。
In order to establish Perilla nankinensis in vitro system of blossom, using the sterile seedlings as the explants, the most suitable condition in vitro blossom were found by the use of hormone and nutrient levels of culture medium, the low temperature and PEG treatment. The results showed that the optima medium for flowering was 6-BA 1.0 mg·L^-1and IAA 0.5 mg·L^-1. The low temperature dealing with 2 days for 10 ℃ was more appropriate for flowering. Flowering would be affected by different nutritional levels.It is favorable to halve the element MS medium. It was helpful to improve the efficiency and quality of flowering by the medium containing with 10%PEG-6000.
