为检测卵泡内小肽的表达,采用基质辅助激光解离串联飞行时间质谱(MALDI-TOF/TOF MS)建立了成熟卵泡液的多肽表达模型,比较小卵泡(直径〈4mm)、中等卵泡(直径4~8mm)和成熟卵泡(直径〉8mm)的多肽表达变化。结果表明,3种形态的卵泡液存在12个显著差异多肽,其中在成熟卵泡内上调6个,下调有5个;在中等卵泡内上调4个,下调5个。其中分子质量约为1746u的多肽在成熟卵泡中特异性表达,可作为卵泡成熟的潜在生物标志物之一。
In order to detect the peptides expression profile in follicles,the matrix-assisted laser desorption ionization mass spectrometry was used to establish a model of peptide expression profile and compare expression changes of peptides from small follicles((φ〈4 mm),medium follicules(4 to 8 mm) and mature follicles(φ〉8 mm).The results indicated that there were 12 peptides to express significantly different among three shapes of follicles.6 peptides increased and 5 peptides decreased in mature follicles.The 1746 u of peptide expressed specifically in mature follicles,which might be used as the one of the underlying biomarkers for follicles maturation.
