通过ELISA测定犬新孢子虫感染对小胶质细胞中细胞因子TNF-α、iNOS、IL-10和Arg-1的影响,鉴定小胶质细胞的活性;用Western blot检测分析犬新孢子虫对小胶质细胞中PPARγ/NF-κB信号通路的影响;探讨犬新孢子虫感染对小胶质细胞活性的影响及机制。结果表明:犬新孢子虫感染5d后,小胶质细胞中IL-10和Arg-1的表达显著升高(P〈0.01),TNF-α和iNOS的表达显著降低(P〈0.01)。PPARγ抑制剂GW9662预处理后,结果则与其相反。同时,犬新孢子虫感染后,能够上调PPARγ的活性,降低p65的磷酸化水平。犬新孢子虫感染能够激活PPARγ/NF-κB信号通路诱导小胶质细胞活性的变化。
The expressions of TNF-a, iNOS, IL-10 and Arg-1 in microglia were determined by ELISA, and then the activity of mieroglia was detected; The activities of PPARv and NF-κB were detected by Western blotting. The aim of this study was to investigate the activity and mechanism of microglia during Neospora caninurn infection. The results showed that the expressions of IL-10 and Arg-I in microglia significantly increased 5 days after the infection (P〈0.01), and the expressions of TNF-and iNOS decreased significantly (P〈0.01). After treatment with the GW9662 inhibitor PPAR, the results were reversed. At the same time, the activity of PPARF increased while the phosphorylation level of p65 decreased. The activity of microglia was changed by activating PPARγ/NF-κB Signaling Pathways during Neospora cani- num infection.
