目的观察人类软骨糖蛋白-39(HCgp-39)对成人膝关节软骨祖母细胞增殖的影响。方法取正常及OA关节软骨标本,向分离培养所得的软骨祖母细胞中分别加入0、25、50、100、500 ng/m L的HCgp-39。收集培养0、24、48、72 h的细胞,加入10μL的MTS检测液,酶标仪检测各孔光的吸收值(OD值),比较各组细胞对HCgp-39的敏感性。结果 OA组:48 h时加入25、50、100、500 ng/m L组的OD值分别为0.81±0.01、0.88±0.02、0.93±0.01、1.03±0.04;72 h分别为1.02±0.05、1.12±0.01、1.21±0.01、1.28±0.01;与未加入HCgp-39组比较均有统计学差异(P<0.05或<0.01);正常软骨组:48 h时加入25、50、100、500 ng/m L组的OD值分别为0.67±0.01、0.63±0.01、0.61±0.01、0.58±0.01;72 h分别为1.18±0.02、1.1±0、1.08±0.01、0.97±0.02;与未加入HCgp-39比较均有统计学差异(P<0.05或<0.01)。结论在成人的关节软骨中存在软骨祖母细胞;HCgp-39能够促进关节软骨中软骨祖母细胞的增殖。
Objective To investigate the effect of HCgp-39 on adult articular cartilage precartilagincus stem cells proliferation.Methods HCgp-39(0, 25, 50, 100, 500 ng/mL) were added into precartilaginous stem cells .After 0, 24, 48, 72 h, stem cells were collected, and subjected to 10μL MTS, and then detecting OD490, comparing the cell sen-sibility to HCgp-39 in all groups.Results OA group:After 24 hours, the mean and standard deviation of cells subjected to 25, 50, 100, 500 ng/mL HCgp-39 were 0.81 ±0.01, 0.88 ±0.02, 0.93 ±0.01, 1.03 ±0.04, separately; after 72 hours, the mean and standard deviation were 1.02 ±0.05, 1.12 ±0.01, 1.21 ±0.01, 1.28 ±0.01, separately.When Compared with stem cells without subjecting to HCgp-39 , there was no statistically differences; normal group: after 24 hours, the mean and standard deviation were 0.67 ±0.01, 0.63 ±0.01, 0.61 ±0.01, 0.58 ±0.01;after 72 hours, the mean and standard deviation were 1.18 ±0.02, 1.1 ±0, 1.08 ±0.01, 0.97 ±0.02;comparing with cells without subjec-ting to HCgp-39, the differences were statistically significant .Conclusions There are articular cartilage cell proliferation in the adult articular cartilage;HCgp-39 had the promotional effects on proliferation of precartilaginous stem cells .
