雌激素对人脐带来源间充质细胞骨架和超微结构的影响

目的研究在17β-雌二醇(E2)作用下人脐带间充质干细胞骨架、超微结构的形态学变化,并分析其骨架蛋白的表达情况及其可能的机制。方法用三酶法分离培养人脐带间充质干细胞,流式细胞仪检测所得细胞表型。E2处理48h后,用激光共聚焦显微镜观察肌动蛋白和纽蛋白的表达情况;用透射电子显微镜观察细胞超微结构的变化。结果分离得到的人脐带间充质干细胞纯度达到95%以上;E2对肌动蛋白的形成和分布没有影响,但使纽蛋白的分布集中于核周;在E2作用下,人脐带间充质干细胞出现老化,自噬体增多,线粒体出现代偿性增加。结论 E2能改变人脐带间充质干细胞的细胞骨架和超微结构,从而可能影响其功能。

英文摘要:

Objective To investigate the effects of 17β-Estradiol (E2) on cytoskeleton and ultrastructure of human umbilical cord mesenchymal stem cells (MSC).Methods The MSC from human umbilical cord were isolated and cultured,followed by identification of immunophenotypes with flowcytometry.The MSC were treated with E2 for 48 hours at 10-7 nmol/L concentrations.Cytoskeleton and ultrastructure of MSC was then observed by laser confocal microscope (LCM) and transmission electron microscope (TEM),respectively.Results Isolated MSC are identified to have over 95% purity.Ageing-like changes and increased autophagosomes and mitochondria are observed in most MSC after treatment with E2.Vinculin aggregates around the nucleus of MSC upon E2 treatment.Conclusion E2 can alter cytoskeleton and ultrastructure of MSC derived from human umbilical cord,thus may influence the function of MSC.
