为研究在基于氨乙酰丙酸(ALA)的光动力治疗(PDT)法中添加适当N掺杂纳米TiO2-xNx对体外灭活白血病HL60细胞的效果影响,采用MTT分析法测试了各种实验条件下HL60细胞的相对存活率,从而研究了纳米TiO2-xNx对HL60细胞生长的影响、添加不同浓度的纳米TiO2-xNx后PDT对HL60细胞存活的影响以及基于ALA或TiO2-xNx或ALA和TiO2-xNx的PDT对HL60细胞的存活影响。实验结果表明:50~250μg/mL浓度的纳米TiO2-xNx溶液对HL60细胞具有较强的灭活作用,其中150μg/mL浓度的纳米TiO2-xNx溶液对任意接种浓度的HL60细胞均具有很好的灭活效果。添加纳米TiO2-xNx再进行PDT,HL60细胞的相对存活率均低于同浓度纳米TiO2-xNx颗粒单独作用后HL60细胞的相对存活率。在ALA的基础上添加纳米TiO2-xNx后进行光辐照,对HL60细胞的灭活效果最佳,当添加ALA浓度为1mM/mL、纳米TiO2-xNx浓度为150μg/mL和HL60细胞接种浓度为7.5×100/mL时,HL60细胞的存活率为25%,即基于添加合适浓度的ALA及纳米TiO2-xNx的PDT对HL60细胞的灭活效率达75%。
In order to study the effects of nanoparticle TiO2-xNx on the inactivation of Leukemic HL60 cells in vitro by use of 5-aminolevulinic acid(ALA) based on photodynamic therapy(PDT), the 3-(4,5-dimethyll-2-thiazolyl)-2-5diphenyl-2H- tetrazolium bromide(MTT) assay was used to evaluate the effects of nanoparticle TiO2-x Nx on the growth of Leukemic HL60,the effects of nanoparticle TiO2-x Nx with different concentrations on the relative survival rate of Leukemic HL60 after PDT and the relative survival rate of Leukemic HL60 based on ALA and PDT(ALA-PDT) by adding nanoparticle TiO2-x Nx. The results indicated that the nanoparticle TiO2-xNx had an obvious inactivation to Leukemic HL60 cells when its final concentration was between 50 μg/mL and 250 μg/mL and the 150 μg/mL was the best concentration especially among them. ALA-PDT based on a suitable concentration of nanoparticle TiO2-xNx could improve the efficiency of inactivation of Leukemic HL60 cells the relative survival rate of Leukemic HL60 cells was the least after PDT based on both ALA and TiO2-xNx.
