目的 探讨转化生长因子(TGF)β1在人涎腺黏液表皮样癌高、低转移细胞株的差异表达。方法 应用基因芯片技术、反转录(RT)-PCR、免疫组化、免疫印迹杂交等方法从基因水平和蛋白水平检测TGF-β1的表达。结果 基因芯片技术、RT-PCR、免疫组化、Western blot结果都证实TGF-β1在人涎腺黏液表皮样癌脊髓转移株高表达,在低转移株不表达。结论 TGF-β1的表达上调可能与人涎腺黏液表皮样癌转移有关。
Objective To investigate the expression of transforming growth factor beta 1 (TGF-β1) in salivary gland mucoepidermoid carci- noma high and low metastatic cell lines. Methods TGF-β1 was examined by gene chip technique, RT-PCR, immunohistoehemistry and Western blot in salivary gland mueoepidermoid carcinoma low metastatic cell line MEC-1 and high metasfie cell line Ms. Results Stronger TGF-β1 expression was observed in Ms cells than in MEC-1 cells by gene chip technique and conformed by RT-PCR, immunohistochemistry and Western blot. Conclusion Over expression of TGF-β1 may contribute to the metastasis of salivary gland mucoepidermoid carcinoma.
