目的:从肺癌组织中纯化并鉴定热休克蛋白70(HSP70),为研究其免疫原性和作为肿瘤疫苗奠定基础。方法:用组织蛋白裂解液从肺癌组织中提取总蛋白,应用ConA-sepharose亲和层析和快速蛋白液相色谱系统(FR-LC)连续分离、梯度洗脱获得蛋白,应用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳及Western-blot进行蛋白相对分子质量及免疫原性鉴定。结果与结论:所获蛋白的相对分子质量为70000,经Western-blot鉴定为HSP70。
Aim: To separate and purify heat shock protein 70 (HSP70) from lung cancer tissue in order to study its immunogenecity and make it a new type tumor vaccines. Methods:The whole soluble proteins of lung cancer tissue were extracted and separated by ConA-sepharose affinity column chromatography and fast protein liquid chromatography system. The relative molecular weight and the immunogenecity of the obtained protein were confirmed by SDS-PAGE and Western-blot. Results and Conclusion: The obtained protein was confirmed to be HSP70 with relative .molecular weight of 70 000.
