目的研究β-榄香烯对人肺腺癌A549细胞株裸鼠移植瘤的放射增敏效应。方法采用细胞悬液接种法建立裸鼠移植瘤模型;将达到0.8~1.0 cm3瘤体积的裸鼠随机分组:空白组(NaCl)、25 mg/kg药物组、45 mg/kg药物组、100 mg/kg药物组、放疗组(NaCl+放疗)、25 mg/kg+放疗组、45 mg/kg+放疗组、100 mg/kg+放疗组,每组5只。应用免疫组化染色检测各组VEGF和CD34的表达水平。结果成功建立裸鼠移植瘤模型,观测各组移植瘤体积倍增时间,计算放射增敏系数,获得最佳增敏剂量45 mg/kg。免疫组化显示药物组VEGF表达水平略低于空白组(P〉0.05);放疗组VEGF和CD34表达明显增强,联合组VEGF和CD34表达明显减弱,均有统计学意义(P〈0.05)。结论β-榄香烯和放疗协同,通过抑制VEGF表达,改善肿瘤血管形成情况,增强了肿瘤细胞的放射敏感性。
Objective The radiosensitive change in nude mice model transplant ed lung adenocarcinoma cell A549 treated by β-elemene was studied,and the associa tion between the possible radiosensitive mechanism and angiogenesis of nude mice model was investigated.Methods The transplanted mice model was established thr ough cell suspension inoculation.The transplanted mice with 0.8~1.0 cm3 tumor were randomized(5 per group):the control(NaCl),25 mg/kg β-elemene,45 mg/kg β-elemene,100 mg/kg β-elemene,radiation(radiation+ NaCl),25 mg/kg β-elemene+radiation,45 mg/kg+ radiation,100 mg/kg β-elemene+ radiation.The expression of VEGF and CD34 in different group was detected by immunohistochemistry.Results The nude mice model was successfully established and the volume double time was obtained and the optimal dose to enhance the radiosensitivity of transplanted tumor was 45 mg /kg.The expression of VEGF and CD34 slightly decreased in β-elemene group(P 0.05),significantly increased in radiation group and apparently decreased in cotreat ment group(P0.05)compared with control group by immunohistochemistry.Conclusion β-elemene has definite activity to enhancing the tumor radiosesitivity and the study indicated it was possible that β-elemene exert the effect by inhibiting the expression of VEGF and CD34.Inhibiting angiogenesis is possibly one of the radio sensitive mechanisms.
