目的:观察不同微弧氧化处理时间对纯钛表面形成的掺锶羟基磷灰石涂层表面性状以及对成骨细胞附着和细胞形态的影响。方法:将抛光后的医用纯钛用微弧氧化法制备,微弧氧化处理时间分别为5、10、15min,掺锶原子数百分比[Sr/(Sr+Ca)]为14%的涂层,分别采用扫描电镜观察表面形貌,用粗糙度仪和接触角测量仪测量各组涂层表面粗糙度和液体接触角;将各组试件与成骨细胞共同培养,用MTT方法检测培养30、60min和90min时成骨细胞在涂层表面的附着情况,并用扫描电镜观察培养3d后的细胞形态。结果:随着微弧氧化时间的延长,涂层表面形貌越不规则,且粗糙度增加,表面水的接触角变小;与成骨细胞共同培养30min时,3组涂层的细胞附着程度无明显差异(P〉0.05),而共同培养60min、90min时,微弧氧化10min和15min组的细胞附着明显高于5min组(P〈0.05);共同培养3d后,成骨细胞在3种涂层表面的附着和伸展均良好,与抛光纯钛相比立体感更强,分泌颗粒较多。结论:通过延长微弧氧化时间可以改变涂层表面特性,并能促进细胞在其表面的早期附着和生长。
AIM:To examine the different surface properties made by micro-arc oxidation(MAO) through different oxidation time,and the effects of these films on osteoblast attachment and cellular morphology.METHODS:Sr-HA coating discs containing 14% of Sr were manufactured by MAO with different oxidation time(5,10 and 15min).The surface morphology were observed by scaning electron microscope(SEM),surface roughness and contact angle were measured using a surface roughness measuring system and a contact angle measuring system.Osteoblast cell adhesion was assayed by MTT method and cellular morphology was observed by SEM.RESULTS:With the oxidation time extended,the surface morphology was much more irregular,surface roughness was enhanced and water contact angle became smaller.Cell culture experiments showed an enhanced osteoblast adhesion on the films formed by 10min and 15min oxidation,and morphological studies by SEM showed a good adhesion behavior of osteoclasts on all three films.CONCLUSION:Extending the oxidation time results in the films to alter surface properties,which benefit osteoblast adhension and growth.
