目的:观察活血补肾方及其拆方对退变腰椎间盘细胞Fas表达和凋亡率的影响。方法:采用纤维环针刺法复制腰椎间盘退变动物模型。造模成功后,将50只新西兰大白兔随机分为活血补肾方、活血方、补肾方、吲哚美辛及模型对照组5组,给药6周。采用流式细胞仪检测椎间盘细胞凋亡率,免疫组化法检测椎间盘细胞Fas表达情况。结果:活血补肾方及其拆方均具有抑制Fas表达并减少椎间盘细胞凋亡的作用。结论:活血补肾方及其拆方抑制Fas表达并减少椎间盘细胞凋亡可能是其治疗椎间盘源性腰痛的机理之一。
Objective:To observe the effects of Huoxue Bushen Decoction and its decomposed formulas on Fas expression and apoptotic rate in degenerated lumbar intervertebral discs.Methods: After establishment animal model of degenerative lumbar intervertebral discs successfully by anulus puncture,50 New Zealand rabbits were randomized divided into Huoxue Bushen Decoction group,Huoxue Decoction group,Bushen Decoction group,Indometacin group,and the model control group.Then each group was given the corresponding medicines for 6 weeks respectively.Flow cytometer technique was used to detect the apoptotic rate of degenerated discs,and enzyme-linked immunosorbent assay method was used to detect Fas expression.Results:Huoxue Bushen Decoction and its decomposed formulas could inhibit the expression of Fas,and reduce the apoptotic rate of the degenerated discs.Conclusion:It was considered that Huoxue Bushen Decoction and its decomposed formulas could significantly inhibite the expression of Fas and depress the apoptotic rate,which could potentially reduce discogenic low back pain.
