目的:观察活血补肾方及其拆方对退变椎间盘组织IL-1β、TNF-α mRNA表达的影响。方法:采用纤维环针刺法复制腰椎间盘退变动物模型。造模成功后,将50只新西兰大白兔随机分为活血补肾方、活血方、补肾方、吲哚美辛及模型对照组5组,给药6周。采用逆转录聚合酶链反应检测IL-1β和TNFαmRNA表达情况。结果:与模型组比较,活血补肾方、补肾方、活血方和吲哚美辛组IL-1β、TNFα mRNA的表达均下调(P〈0.01)。结论:活血补肾方具有抑制IL-1β、TNFα mRNA表达的作用,这可能是活血补肾方减轻退变椎间盘炎症反应,减缓椎间盘退变,治疗椎间盘源性腰痛的机理之一。
Objective: To observe the effects of Huoxue Bushen Decoction and its decomposed formulas on expression of IL-1β and TNF-α mRNA in degenerated lumbar intervertebral discs.Methods: After establishment animal model of degenerative lumbar intervertebral discs successfully by anulus puncture,50 New Zealand rabbits were randomized divided into Huoxue Bushen Decoction group,Huoxue Decoction group,Bushen Decoction group,Indometacin group,and the model control group.Then,the expression of IL-1β and TNF-α mRNA in degenerated lumbar intervertebral discs were detected by RT-PCR after treated for 6 weeks.Results: The expression of IL-1β and TNF-α mRNA in the model control group were higher compared with the other four groups respectively(P〈 0.01).Conclusion: It was considered that Huoxue Bushen Decoction and its decomposed formulas could significantly inhibite the expression of IL-1β and TNF-α mRNA,which could potentially decrease the inflammation reaction,delay degeneration of lumbar intervertebral discs,and reduce discogenic low back pain.
