目的 分析Bmi-1与Ki-67,Bcl-2在结直肠癌中表达之间的关系,探讨其与结直肠癌临床病理因素的关系及临床意义.方法 采用免疫组织化学PV-9000法检测84例结直肠癌及配对正常肠黏膜,21例结直肠上皮内肿瘤中的Bmi-1、Ki-67和Bcl-2蛋白的表达.结果 Bmi-1在结直肠癌(73.81%)、上皮内肿瘤(95.23%)中的阳性表达率显著高于正常肠黏膜(59.52%),P<0.05.Bmi-1在结直肠癌中的表达与结直肠癌的WHO组织学分型、Dukes分期和淋巴结转移相关,P值均>0.05.在84例结直肠癌中Bmi-1的表达与Bcl-2的表达呈正相关,等级相关系数r=0.358,P<0.001.结论 结直肠癌和结直肠上皮内肿瘤中Bmi-1蛋白的异常表达可能与细胞增殖、凋亡和癌变有关.Bmi-1和Bcl-2在结直肠癌中的表达密切相关,表明Bmi-1可能协同Bcl-2发挥抗凋亡效应,但二者抑制凋亡途径不同.
Objective To analyze the correlation between Bmi - 1, Ki -67 and Bcl -2 expression in colorectal cancer and to explore the correlation and significance with clinicopathological features of colorectal cancer. Methods Immunohistochemistry staining was used to display the expression of Bmi - 1, Ki - 67 and Bel -2 in 84 cases of coloreetal cancer,84 cases of matched normal colorectal mueosa and 21 cases of precancerous lesion. Results The positive rates of Bmi - 1 expression in colorectal cancer (73.81% ) and intraepithelial neoplasia(95.23% )were significantly higher than those in normal colorectal mueosa membrane (59.52%), P 〈 0.05. There was a positive relationship between Bmi - 1 and Bcl - 2 expression in 84 cases of colorectal cancer( r = 0.358, P 〈 0.001 ). Conclusion The abnormality of Bmi - 1 expression in coloreetal cancer might be involved in cell proliferation,apoptosis and cancerization. Bmi -1 was closely correlated with Bc1-2 in eoloreetal cancer, suggesting that Bmi - 1 might coordinate with Bel - 2 to induce anti - apoptosis effect, whereas the suppressing apoptosis pathways were different.
