目的探讨氧糖剥夺对星形胶质细胞内Cdh1蛋白表达的影响及其机制。方法①体外纯化培养大鼠大脑皮层星形胶质细胞,随机分为对照组、氧糖剥夺1h复氧组、氧糖剥夺6h复氧组,Western blot检测Cdh1及Skp2蛋白的表达变化;②将体外纯化培养的星形胶质细胞随机分为对照组、氧糖剥夺6h组、单纯缺氧6h组,Western blot检测Cdh1蛋白的表达变化,血糖仪检测培养液中葡萄糖含量的变化。结果①与对照组相比,氧糖剥夺1h复氧组、氧糖剥夺6h复氧组Cdh1蛋白表达均下降,Skp2蛋白表达均增加(P〈0.05),氧糖剥夺1h复氧组与氧糖剥夺6h复氧组组间Cdh1、Skp2蛋白表达差异无统计学意义;②与对照组相比,氧糖剥夺6h组Cdh1蛋白表达明显下降,单纯缺氧6h组没有明显变化,氧糖剥夺6h组与单纯缺氧6h组组间差异有统计学意义(P〈0.05);③单纯缺氧6h组细胞外液葡萄糖摄取率低于对照组(即常氧组)[(21.43±6.74)%vs.(26.98±9.21)%,P〈0.05]。结论氧糖剥夺后星形胶质细胞内Cdh1蛋白表达减少,其变化与细胞外液中缺少葡萄糖有关。

Objective To explore the influence of oxygen-glucose deprivation(OGD)on Cdh1 protein expression in vitro cultured astrocytes and the mechanism.Methods The cerebral cortex astrocytes of rats were purified and cultured in vitro,and randomly divided into the control group,OGD 1 h and recovery(OGD 1 h/R)group,OGD 6 h and recovery(OGD 6 h/R)group.The expression of Cdh1 and Skp2 proteins was detected by using Western blot.Astrocytes were randomized into control group,OGD 6 h group and hypoxia 6 h group,and the expression of Cdh1 protein was detected by using Western blot.Blood glucose meter was used to test glucose change.Results ① As compared with control group,the expression of Cdh1 in OGD 1 h/R group and OGD 6 h/R group was decreased and the expression of Skp2 was increased(P0.05),but there was no statistically significant difference between OGD 1 h/R group and OGD 6 h/R group;② As compared with control group,the expression of Cdh1 in OGD 6 h group was significantly decreased,and there was statistically significant difference between OGD 6 h group and hypoxia 6 h group(P0.05).Glucose uptake rate of extracellular fluid in hypoxia 6 h group was lower than in control group[(21.43±6.74)% vs.(26.98±9.21)%,P0.05].Conclusion The expression of Cdh1 in astrocytes after OGD was decreased,and the change was related to the lack of glucose in extracellular fluid.
