目的研究新型褪黑素(melatonin,MLT)受体激动剂Neu-P11对睡眠限制大鼠胰岛素敏感性的影响。方法采用转笼法对SD大鼠进行8d的睡眠限制,期间每天分别给予腹腔注射Neu-P11(20mg·kg^-1)、MLT(5mg·kg^-1)、生理盐水。监测各组大鼠体重和耗食量,实验末测定空腹血糖、胰岛素、TG、TC、HDL-C水平,进行口服葡萄糖耐量实验(OGTT)。结果睡眠限制后大鼠空腹血糖、血清胰岛素和TG、TC水平明显高于正常对照组,HDL-C水平及葡萄糖耐量降低;而与睡眠限制组比较,Neu-P11或MLT处理组血糖、血清胰岛素、TG、TC水平及HOMA-IR降低、葡萄糖耐量升高。结论Neu-P11能调节睡眠限制大鼠的糖脂代谢,改善睡眠限制引起的胰岛素抵抗。
Aim To investigate the influence of Neu- P11, a novel melatonin receptor agonist, on insulin sensitivity in sleep restricted rats. Methods Method of rotating cage was adopted to establish SD rat models of sleep restriction. During the 8 days of sleep restriction, rats were injected intraperitoneally with Neu-P11 (20 mg· kg^- 1)/ MET (5 mg· kg^-1)/ saline respectively every day. Body weight and food consumption were monitored every day. Plasma glucose, fasting insulin, TG, TC, HDL-C levels were detected while the oral glucose tolerance test (OGTT) was performed at the end of experiment. Results Compared with control group, sleep restricted rats exhibited an increase of plasma glucose, fasting insulin, TG and TC, whereas HDL-C and glucose tolerance decreased obviously. However, compared with sleep restriction rats treated by vehicle, the levels of plasma glucose, TG, TC and HOMA-IR decreased with an increase of HDL-C and glucose tolerance in Neu-P11 or melatonin-treated sleep restricted rats. These data suggested that the metabolism and insulin sensitivity of the chronic sleep restricted rats were protected by Neu-P11 as well as melatonin. Conclusion Neu-P11 could improve metabolic profiles and insulin sensitivity, and improve insulin resistant induced by sleep restriction.
