目的:探讨SLE患者外周血单个核细胞(PBMCs)BLyS基因和蛋白表达及糖皮质激素地塞米松对其表达的影响。方法:梯度密度离心法分离25例SLE患者[(31.40±14.23)岁]和20名女性健康志愿者[(28.20±10.36)岁]的PBMCs,分为2组,地塞米松(1μmol/L)组和培养基组(仅含RPMI一1640培养基),分别于培养0h、6h、12h、24h和72h离心收集PBMCs,用RT—PCR法检测刺激后0至24h时点细胞BLyS mRNA的表达情况,流式细胞仪(FACs)和直接免疫荧光法(激光共聚焦荧光显微镜)检测72h膜结合型BLyS蛋白的表达。结果:(1)SLE患者BLyS mRNA和蛋白表达均高于健康对照(0.40±0.18vs0.27±0.20,P〈0.01;0.37±0.17vs0.25±0.17,P〈0.01;0.42±0.26vs0.29±0.16,P〈0.01;0.39±0.23vs0.24±0.18,P〈0.01)。(2)地塞米松显著抑制健康对照和SLE患者PBMCs BLyS mRNA表达,12h最为明显(0.19±0.10vs0.31±0.16,P〈0.01;0.29±0.18vs0.46±0.21,P〈0.01)。(3)地塞米松显著抑制健康对照和SLE患者PBMCs膜结合型BLyS蛋白表达(2.73±1.89vs3.67±1.64,P〈0.01;3.42±1.53vs4.52±1.95,P〈0.01)。(4)直接免疫荧光法激光共聚焦荧光显微镜检测PBMCs膜结合型BLyS蛋白表达显示地塞米松刺激后BLyS表达减弱。结论:SLE患者PBMCsBLyS表达增强,其表达受地塞米松负影响。
AIM : To determine the expression of membrane - bound B lymphocyte stimulator (BLyS) and its mRNA in peripheral blood mononuclear cells (PBMCs) from individuals with systemic lupus erythematosus (SLE), and to investigate the effect of dexamethasone on BLyS expression. METHODS: PBMCs were obtained from 25 individuals with SLE ( mean age of 31.40 ± 14. 23 ) and 20 female healthy volunteers ( mean age of 28.20 ± 10. 36). They were randomized into dexamethasone ( 1 μmol/L) group and media group. PBMCs were gathered at 0, 6, 12 and 24 h for BLyS mRNA assessment using reverse transcription - PCR ( RT - PCR). PBMCs were also collected at 72 h for membrane - bound BLyS protein detection using flow cytometry (FACS) and direct immunofluorescence. RESULTS: (1) The expression of BLyS mRNA and membrane -bound protein were significantly higher in PBMCs from individuals with SLE than that in PBMCs from healthy controls (0.40±0. 18 vs 0.27±0.20, P〈0.01; 0.37 ±0. 17 vs 0.25 ±0. 17, P〈0.01; 0.42 ±0.26 vs 0. 29 ± 0. 16, P 〈 0.01 ; 0. 39 ± 0. 23 vs 0. 24 ± 0. 18, P 〈 0. 01 ). ( 2 ) Dexamethasone suppressed BLyS mRNA expression in PBMCs from both healthy controls and individuals with SLE, with the greatest effect at 12 h (0. 19 ±0. 10 vs 0. 31 ± 0. 16, P 〈 0. 01 ; 0. 29 ± 0. 18 vs 0. 46 ± 0. 21, P 〈 0. 01 ). (3) Dexamethasone also inhibited the expression of membrane - bound BLyS protein in PBMCs from both healthy controls and individuals with SLE ( FACs, mean fluorescence intensity, 2.73 ± 1.89 vs 3.67± 1.64, P 〈0. 01 ; 3.42 ± 1.53 vs 4. 52±1.95, P 〈0. 01 ). (4) Direct immunofluorescence by confocus micrography also indicated that dexamethasone decreased the expression of membrane - bound BLyS protein.CONCLUSION: In vitro, the expression of BLyS mRNA and membrane - bound protein is elevated in individuals with SLE, and dexamethasone has negative effects on its expression.
