目的探讨系统性红斑狼疮(SEE)患者外周血单个核细胞(PBMCs)B淋巴细胞刺激因子(BLys)基因和蛋白在体外的表达及干扰素(IFN)-γ对其表达的调控。方法梯度密度离心法分离25例SLE患者[(31±14)岁]和20名健康志愿者[(28±10)岁]的PBMCs,分为2组,IFN-γ(5ng/m1)组和培养基组(仅含1640培养基),分别于培养0、6、12、24和72h离心收集PBMCs,用反转录聚合酶链反应(RT—PCR)法检测刺激后0~24h时点细胞BLySmRNA的表达。流式细胞仪(FACs)和直接免疫荧光法(激光共聚焦荧光显微镜)检测72h膜结合型BLyS蛋白的表达。结果(1)SEE患者PBMCsBLySmRNA和蛋白的表达在体外均强于健康对照(P〈0.05);②IFN-γ显著增强健康对照和SLE患者PBMCsBLySmRNA表达,6h最为明显(0.42±0.19vss0.25±0.14,P〈0.01;0.59±0.28vs0.44±0.21,P〈0.01);③IFN-γ可显著增强健康对照和SLE患者PBMCs膜结合型BLyS蛋白表达(4.5±3.0vs3.7±2.6,P〈0.01;5.7±3.81vs4.5±2.9,P〈0.01);④直接免疫荧光法激光共聚焦荧光显微镜检测PBMCs膜结合型BLyS蛋白表达提示IFN-γ刺激后BLyS表达增强。结论SLE患者PBMCsBLyS表达增强,而IFN-γ可增强患者BLyS基因和蛋白表达。
Objective To determine the expression of membrane-bound B lymphocyte stimulator (BLyS) protein and its mRNA in vitro of peripheral blood mononuclear cells (PBMCs) from individuals with systemic lupus erythematosus (SLE), and to investigate the role of interferon-γ (IFN-γ) on the expression of BLyS. Methods PBMCs were obtained from 25 SLE patients (mean age of 31±14) and 20 healthy volunteers (mean age of 28±10). They were randomized into IFN-γ (5 ng/ml) group and control group. PBMCs were collected at 0, 6, 12 and 24 h for BLyS mRNA assessment using semi-quantitative reverse transcription-PCR (RT-PCR). PBMCs were also collected at 72 h for membrane-bound BLyS protein detection using flow cytometry (FACS) and direct immunofluorescence. Results (1)The expression of BLyS mRNA and membranebound protein in PBMCs was significantly higher in individuals with SLE compared with healthy controls (P〈 0.05); (2) IFN-γ enhanced BLyS mRNA expression in PBMCs in both healthy controls and SLE patients, with the greatest effect at 6 h (stimulated vs unstimulated, 0.42±0.19 vs 0.25±0.14, P〈0.01; 0.59±0.28 vs 0.44± 0.21, P〈0.01 ); (3)IFN-γ also increased the expression of membrane-bound BLyS protein in both healthy controls and individuals with SLE (FACs, mean fluorescence intensity, 4.5±3.0 vs 3.7±2.6, P〈0.01; 5.7±3.8 vs 4.5±2.9, P〈0.01 ); (4)Direct immunofluorescence by con-focus micrography also indicated that IFN-γ increased the expression of membrane-bound BLyS protein. Conclusion In vitro, the expression of BLyS mRNA and membrane-bound protein is elevated in SLE patients, and IFN-γ has positive effects in its expression.
