目的:探讨转录因子7类似物2(transcription factor 7 like 2,TCF7L2)基因rs12255372位点多态性与亚洲人群2型糖尿病(T2DM)发病的相关性。方法:通过电子计算机检索国内外有关TCF7L2基因rs12255372与T2DM关系的病例对照或队列研究文献,提取入选文献相关数据并应用Rev Man 5.3软件进行固定效应及随机效应模型Meta分析。结果:筛选到27篇符合要求的文献,包括T2DM病例15 300例,健康人群对照12 693例;异质性检验显示各研究间存在有统计学意义的异质性(Q=57.10,P=0.002);rs12255372位点等位基因(T/G)的分布频率在病例组和对照组之间差异有统计学意义(P〈0.000 1),病例组的T等位基因分布频率显著高于对照组,合并OR(95%CI)为1.45(1.32,1.59);发表偏倚分析和敏感性分析结果证实,上述Meta分析结果稳定、可靠。结论:rs12255372位点T等位基因可增加亚洲人群T2DM的发病风险,TT/TG基因型是T2DM的风险基因型。
Objective: To evaluate the association of the transcription factor 7-like 2( TCF7L2) polymorphism rs12255372 with type 2 diabetes mellitus( T2DM) in Asian population. Methods: Literature of case-control study or cohort study on TCF7L2 rs12255372 polymorphism with T2 DM in Asian population was collected. To do the meta-analysis,the fixedeffects and random-effects model within the Rev Man 5. 3 software was used. Results: Twenty-seven eligible papers( with15,300 cases and 12,693 controls) were included in the final meta-analysis. The heterogeneity test revealed the significant heterogeneity in the studies( Q = 57. 10,P = 0. 002). The rs12255372 T allele was significantly associated with T2DM( P 〈0. 0001),considering the T allele itself( OR = 1. 45,95% CI = 1. 32,1. 59). Publication bias and sensitivity analysis confirmed the meta-analysis results. Conclusion: It is confirmed that rs12255372 is significantly associated with susceptibility to T2 DM in Asian population,the risk allele and the genotypes will increase the risk of T2 DM.
