目的观察活化的肝星状细胞中Septin4_I2基因的表达,并对其进行生物信息学分析。方法采用实时荧光定量PCR法观察活化的肝星状细胞LX-2中Septin4_I2基因的表达,利用网络平台和生物软件分析Septin4_I2序列。结果活化的肝星状细胞中Septin4_I2基因表达上调。Septin4_I2基因序列含有5个内含子及保守的GTP结合位点。Septin4_I2蛋白以loop结构为主,氨基酸序列中含有数个丝氨酸、苏氨酸及酪氨酸磷酸化位点,含有特异性的蛋白激酶K、蛋白激酶C及酪蛋白激酶Ⅱ磷酸化位点,含有N-豆蔻酰化位点。结论活化的肝星状细胞Septin4_I2基因表达上调,编码蛋白可能与细胞分裂相关。
Objectives To observe Septin4_I2 expression in activated hepatic stellate cells (HSCs) and to sequence Septin4_I2. Methods Septin4_I2 expression in activated HSCs was analyzed with quantitative real-time PCR. The sequence of Septin4_I2 was analyzed with bioinformatics using web-based tools and several programs. Results Septin4_I2 expression was up-regulated in activated HSCs. The Septin4_I2 sequence had five introns and GTP-binding sites were also present in Septin4_I2. The Septin4_I2 protein consists primarily of a loop structure. It is predicted to contain serine phosphorylation sites, threonine phosphorylation sites, tyrosine phosphorylation sites, protein kinase C and protein kinase A phosphorylation sites, casein kinase Ⅱ phosphorylation sites, and N-myristoylation sites. Conclusion The expression of Septin4_I2 is up-regulated in activated HSCs. Septin4_I2 may play an important role in the process of cell division.
