目的研究组分配伍四逆汤对甲状腺功能减退症并发高脂血症及心血管系统损害的影响。方法将96只大鼠随机分为空白组、模型组、阳性对照组、四逆汤有效组分高剂量组(CSDH组)、四逆汤有效组分中剂量组(CSDM组)、四逆汤有效组分低剂量组(CSDL组)。除空白组外,其他组连续灌胃丙硫氧嘧啶(PTU)溶液0.01 g/kg 15 d制造甲状腺功能减退症模型,用组分配伍四逆汤高、中、低剂量和左甲状腺素片(优甲乐)对此动物模型进行治疗,连续给药1个月,治疗同时隔日给予0.1%PTU维持甲状腺功能减退症状态,实验结束后,取血,离心分离血清;取心脏做10%心脏组织匀浆,以血清胆固醇(TC)、三酰甘油(TG)和低密度脂蛋白胆固醇(LDL-C)为指标,观察组分配伍四逆汤对甲状腺功能减退症并发高血脂的影响;以血清心钠素(ANP)、内皮素(ET)及心肌组织Na^+-K^+-ATP酶的活力为指标,观察组分配伍四逆汤对甲状腺功能减退症心血管系统的影响。结果与空白组比较,模型组血清三碘甲状腺原氨酸(T3)、甲状腺素(T4)水平明显降低(P〈0.01),高敏促甲状腺激素(TSH)水平显著升高(P〈0.01);与模型组相比,四逆汤有效组分高、中剂量组可升高血清T3、T4水平,降低TSH水平(P〈0.05);与空白组相比,模型组TG、TC及LDL-C明显升高(P〈0.05或P〈0.01);与模型组相比,四逆汤有效组分高、中剂量组的TG、TC及LDL-C明显减少(P〈0.05)。与空白组比较,模型组及各药物治疗组血清ANP、ET水平及心脏组织Na^+-K^+-ATP酶显著降低(P〈0.05或P〈0.01),与模型组相比,四逆汤有效组分各剂量组ANP水平显著升高(P〈0.01),四逆汤有效组分配伍高剂量组血清ET水平明显升高(P〈0.05),四逆汤有效组分配伍组心脏组织Na^+-K^+-ATP酶有所升高,但差异无统计学意义。结论组分配伍四逆汤可缓解甲状腺功?
Objective To investigate the influences of component comptibility Sini decoction( CCSD) on hypothyroid complicated with hyperlipemia and cardiovascular injury. Methods The hypothyroidism model was established by 0. 1% propylthiouracil( PTU) solution 10 m L / kg for 15 days. The hypothyroidism animal were randomly divided into model group,CCSD high,middle,low dose group and positive drugs group,at the same time the mice should give the 0. 1% PTU for 4 weeks. At the end of the experiment,took the blood and kidney,separated the serum. Radioimmunoassay was used to determine the serum triiodothyronine( T3),thyroxine( T4). UV spectrophotometer was used to detected the total cholesterol( TC),triglyceride( TG),low-density lipoprotein cholesterol( LDL-C),myocardium Na^+-K^+-ATP enzyme activity concentration. ELEASE method were used to measure the serum atrial natriuretic peptide( ANP),endothelin( ET). Results Compared with blank group,T3,T4 concentration were decreased while thyroid-stimulating hormone( TSH) concentration was increased in model group(P〈0. 01). Compared with model group,TG,TC,LDL-C levels were decreased in CCSD high,middle dose group(P〈0. 05). Compared with blank group,TG,TC,LDL-C levels were increased(P〈0. 05 or P〈0. 01). Compared with model group,T3,T4 concentration were increased while TSH concentration was decreased in CCSD high,middle dose group(P〈0. 05). The levels of ANP,ET and Na^+-K^+-ATP enzyme activity in model group and CCSD group(P〈0. 05 or P〈0. 01). Compared with model group,ANP level was increased in CCSD groups(P〈0. 01). ET level was increased in CCSD high dose group(P〈0. 05). Conclusion CCSD can alleviate hyperlipemia and cardiovascular injury in hypothyroidism model rats.
