目的:观察丹参总酚酸对慢性心力衰竭大鼠心肌肌浆网Ca2+-ATP酶(Ca2+-ATPase,SERCA)、受磷蛋白(Phospholamban,PLB)的蛋白表达的影响,探讨丹参总酚酸抗心力衰竭的作用机制。方法:结扎大鼠冠状动脉左前降支造成心肌梗死后慢性心力衰竭模型,连续给予丹参总酚酸4周后,采用蛋白印迹(Western Blot)技术检测心肌组织的SERCA和PLB蛋白表达水平及PLB的磷酸化水平。结果:与心力衰竭模型组相比,丹参总酚酸能明显升高SERCA及磷酸化受磷蛋白(phosphorylated Phospholamban,P-PLB)的蛋白表达及PLB的磷酸化水平。丹参总酚酸能明显降低PLB/SERCA比值。结论:丹参总酚酸能升高SERCA的蛋白表达,升高P-PLB的蛋白表达及PLB的磷酸化水平,降低PLB/SERCA比值,减轻PLB对SERCA功能的抑制,这可能是丹参总酚酸抗心力衰竭的机制之一。
Objective:To observe the effect of Salvianolic acids on the protein expression of Ca2+-ATPase(SERCA),Phos-pholamban(PLB) in chronic heart failure(CHF) rats.Methods:The model of CHF was induced by ligating the anterior descend-ing branch of the left coronary arteryt,hen giving the Salvianolic acids by gavage for four successive weeks.The effect of expres-sion of SERCA,PLB was observed by western blot.Results:Compare with model groupt,he Salvianolic acids could increase the protein expression of SERCAi,ncrease the protein expression of phosphorylated Phospholamban(P-PLB) significantly.The Salvi-anolic acids could increase the Phosphorylation level of PLB,and decrease PLB/SERCA significantly.Conclusion:Salvianolic acids could increase the protein expression of SERCA and P-PLBi,ncrease the Phosphorylation level of PLB,decrease the inhibi-tion of PLB to SERCA functioni,t might be one of the mechanism of Salvianolic acids against heart failure.
