目的综合考察雪菊花茶的质量,建立质量评价方法。方法采用TLC和UPLC法建立雪菊中异奥卡宁葡萄糖苷的定性鉴别法和含量测定法,测定10批样品中异奥卡宁葡萄糖苷含量及水分、总灰分、酸不溶性灰分、水溶性浸出物和醇溶性浸出物等常规理化试验项目。结果 TLC图谱中,在各样品中均可检视出异奥卡宁葡萄糖苷的斑点;UPLC测定法中,异奥卡宁葡萄糖苷色谱峰分离度好,在5.1~40.8 mg·L-1内线性关系良好,回收率为98.7%(RSD1.7%),在10批样品中异奥卡宁葡萄糖苷含量质量分数为0.361~0.681%;常规理化试验项目测定结果表明:各样品中仅酸不溶性灰分值差异较大,其他项目测定值集中分布在平均值±40%区间内。结论建立的TLC和UPLC分析方法及采用的理化试验项目适宜作为雪菊综合质量评价方法。
Objective To systematically evaluate the quality of the flower tea from Coreopsis tinctoria and establish the method for quality control. Methods Isookanin-7-O-β-D-glucopyranoside in 10 samples of the flower tea from Coreopsis tinctoria was examined by TLC and its content in samples was determined by UPLC. Some physic- chemical tests( H_2O,total ash,acid- insoluble ash,H_2O extract and 75 %( φ) Et OH extract) were done to investigate their quality. Results The spots of isookanin-7-O-β-D-glucopyranoside in 10 samples were seen in the TLC chromatogram. A good linearity for isookanin-7-O-β-D-glucopyranoside within the range of 5.1-40.8 mg·L(-1) was showed,and the recovery was 98.7 %( RSD1.7 %). The contents of isookanin-7-O-β-D-glucopyranoside in 10 samples were 0.361-0.681%. The values of physic-chemical parameters expect for acid-insoluble ash were the range of ± 40 % of average value. Conclusions TLC,UPLC and physic-chemical tests used in this study are suitable for the systematical quality control of the flower tea from Coreopsis tinctoria.
