目的 研究不同温度和不同浓度B(a)P-7,8-二醇-9,10-环氧化物(BPDE)作用A549细胞后,热休克蛋白27(HSP2)、热休克蛋白70(Hsp70)的表达规律。方法 体外培养的A549细胞分为高温应激组和BPDE组。37℃培养的细胞作对照,高温应激组细胞置于高温(39℃、42℃、43℃)应激2小时。BPDE组用不同浓度的BPDE(0、2、4、8gmol/L)染毒2h。利用Westem-blot技术分析A549细胞株中Hsp27、Hsp70的表达水平。结果 高温应激组细胞HsD27的表达较37qc对照明显升高,差异均有显著性(P〈0.05)。在39℃,Hsp27的表达水平达到峰值。高温处理后,Hsp70的表达也增加,在42℃达到最高,且与对照相比差异有显著性(P〈0.05)。BPDE组HsD27、Hsp70的表达均较对照明显升高,其中Hsp27的表达水平在实验用最高浓度8μmol/L达到最高,且与对照比较差异有显著性(P〈0.05),但仍处于上升阶段。Usp70在4μmol/L达到最高,在4μmol/L和8μmol/L时,Hsp70的表达水平与对照比较差异有显著性(P〈0.05)。结论 高温和BPDE均能诱导A549细胞Hsp27、Hsp70的表达。高温应激时,Hsp27和Hsp70的表达水平分别在39℃和42℃达到峰值。BPDE处理时,Hsp27在8μmol/L时表达最高,但仍处于上升趋势,而Hsp70在4μmol/L时表达最高。
Objective To investigate the expression of the Hsp27 and Hsp70 in A549 cells treated with heat and BPDE. Methods A549 ceils cultured in vitro were divided into heat stress group and BPDE group. Cells cultured at 37% used as control. The heat stress group was exposed to heat at different temperature(39%, 42% and 43% )for 2h. The BPDE group was treated with different concentrations of BPDE (0, 2, 4 and 8μmol/L). Western-blot was used for the Hsp27 and Hsp70 expression analysis. Results In heat stress group, Hsp27 expression levels significantly increased compared to the control (P 〈 0.05) and reached its peak at 39℃ . Hsp70 expression level also increased after heat stress. It reached the peak at 42% and was higher than that of the control with significance( P 〈 0.05). In BPDE group, Hsp27 and Hsp70 expression levels were higher than those of the control, Hsp27 reached a maximum at 8μmol/L, the highest concentration of BPDE in this experiment, and was significantly higher than that of the control( P 〈 0.05). But it was still on its elevatory phase. Hsp70 reached its peak at 4μmol/L and was significantly higher than that of the control at 4μmol/L and 8μmol/L. Conclusion Both heat and BPDE can induce the expressions of Hsp27 and Hsp70. In heat stress group, the Hsp27 and Hsp70 levels reached their peaks at 39% and 42℃. In BPDE group, the expressional levels of Hsp27 and Hsp70 reached their peaks at 8μmol/L and 4μmol/L.
