目的:探讨穿山龙总皂苷对高尿酸血症的降尿酸作用并通过体外实验验证其抗炎作用。方法:将小鼠分为正常组、模型组、别嘌呤醇组和穿山龙总皂苷高中低剂量组。观察小鼠血清尿酸值(SUA),黄嘌呤氧化酶(XOD)和腺苷脱氨酶(ADA)活性。进一步检测肾组织阴离子转运体1(OAT1)的mRNA表达情况。提取人多形核白细胞(PMNL),药物干预后用微晶型尿酸钠(MSU)进行刺激,观察各组β-葡萄糖醛酸酶(β-GD)和乳酸脱氢酶(LDH)的释放率。结果:穿山龙总皂苷高、中、低(600、300、60mg/kg)剂量均可显著降低高尿酸血症小鼠血清SUA水平,并降低血清XOD和ADA活性(P〈0.05);同时可回调OAT1 mRNA的表达量(P〈0.05)。穿山龙总皂苷高、中、低(200、100、50μg/mL)剂量均可显著抑制与MSU进行孵育的多形核白细胞(PMNL)中β-GD和LDH的释放率(P〈0.05)。结论:穿山龙总皂苷具有降高尿酸血症血尿酸和抗炎作用,其降尿酸的机制可能是通过抑制尿酸生成并促进尿酸的排泄而实现的。
Objective: To study the effect of total saponins from Rhizoma Dioscoreae Nipponicae on hyperuricemia models and verify its anti-inflammatory effect in vitro. Methods: Mice were divided into normal group, model group, positive group and total saponins groups of different doses. SUA as well as the activities of XOD and ADA in the blood serum of hyperudcemia mice were observed. The rnRNA expression of OAT1 in the renal was further identified by realtime PCR. Human poly-morphonuclear leucocytes (PMNL) were extracted. After incubated with drugs, MSU was used to stimulate the cells. Release rates of β-glucuronidase (β-GD) and lactate dehydrogenase (LDH) were observed. Results: Total saponins from Rhizoma Dioscoreae Nipponicae of different doses (600, 300, 30mg/kg) could significantly reduce uric acid level and the activities of both XOD and ADA in the serum of hyperuricemia mice (P〈0.05). Meanwhile, it could up-regulated mRNA expression of OAT1 (P〈0.05). Total saponins from Rhizoma Dioscoreae Nipponicae of different doses (200, 100, 50μg/mL) could significantly inhibit the releasing rates of β-GD and LDH from PMNL which were incubated with MSU (P〈0.05). Conclusion: There were uric acid reducing effect as well as anti-inflammatory effect of total saponins from Rhizoma Dioscoreae Nipponicae. The mechanism of reducing uric acid might be caused by inhibiting the production of uric acid and promoting the excretion of uric acid.
