目的:分析重金属暴露秀丽线虫中咽泵运动与寿命之间的相关性。方法:毒性评估实验选择重金属Hg、Pb、Cr与Cd。分析重金属暴露秀丽线虫中咽泵运动的变化情况,并进一步利用线性回归方法分析重金属暴露秀丽线虫中咽泵运动与寿命之间的相关性。结果:在所有评价浓度下的Hg与Pb暴露以及100与200μmol.L-1浓度下的Cr与Cd暴露均可以显著降低咽泵的运动速率。咽泵运动速率的变化显著相关于秀丽线虫所暴露重金属的浓度。而且,重金属暴露秀丽线虫中的咽泵运动速率显著相关于动物的寿命长度。结论:重金属暴露秀丽线虫中存在咽泵运动速率与寿命长度之间的显著关联,且在分析环境有害物质的毒害时,其对于秀丽线虫寿命的毒害可以通过咽泵运动速率来间接进行评价。
Objective:To investigate the association of pharyngeal pumping with mean lifespan in metal exposed nematode Caenorhabditis elegans.Methods:The metals of Hg,Pb,Cr,and Cd were selected for toxicity assay.The alterations of pharyngeal pumping in metal exposed nematodes were examined.In addition,the association of pharyngeal pumping with mean lifespan in metal exposed nematodes was determined by linear regression method.Results:Exposure to Hg and Pb at all examined concentrations and to Cr and Cd at concentrations from 100 μmol·L-1 to 200 μmol·L-1 caused a significant decrease of pumping rate.The alterations of pharyngeal pumping rate in metal exposed nematodes were significantly correlated with the concentrations of examined metals.Moreover,the pharyngeal pumping rate was significantly correlated with mean lifespan in metal exposed nematodes.Conclusion:The close association of pharyngeal pumping with mean lifespan can be observed in metal exposed nematodes,and the endpoint of lifespan can be effectively substituted by the endpoint of pumping rate while assessing the toxicity on aging from environmental toxicants.
