为进一步揭示显性核不育亚麻不育分子机理,以显性核不育亚麻不育株分离的可育花蕾和不育花蕾为研究材料,采用实时定量PCR技术和原位杂交技术,开展了G-E07-830、G-E07-330和G-E07-100这3个花蕾发育相关基因在亚麻花蕾中的时空表达模式。结果表明:原位杂交中G-E07-830、G-E07-330和G-E07-100基因在亚麻可育和不育花蕾绒毡层中表达特异性信号;实时定量PCR得出在早期不育亚麻花蕾中表达量显著大于其他发育时期的花蕾。
In this paper,the real time quantitative PCR technology and in situ hybridization method were adopted,temporal and special expression of three candidate genes G-E07-830,G-E07-330 and G-E07-100 in fertile and sterile flower buds separated from sterile flax of dominant genomic male sterile flax. The results showed that the three genes were specifically expressed in the tapetum of fertile and sterile flower buds. They were expressed at significantly higher levels in the early stage sterile buds than other developmental stages. The results provides some valuable information for the revealing of the molecular mechanism of pollen development and male sterility in flax.
