亚麻雄性不育相关基因MS2-F在亚麻花粉发育过程中起重要的作用。为了研究其功能,构建了2个RNAi载体。选定该基因的2个不同的片段,分别以正向和反向插入到pHANNIBAL载体中PDK intron的两侧,以形成hpR-NA表达盒。然后将hpRNA表达盒插入到pART27的NotⅠ酶切位点上,成功构建了2个亚麻MS2-F基因的RNAi载体。最后将载体导入到农杆菌LBA4404中。RNAi载体成功构建,为验证MS2-F基因在亚麻花粉发育过程中的生物学功能奠定基础。
Flax male sterility related MS2-F gene plays an important role in the process of flax pollen development.In order to conform the function of this gene,two RNAi vectors of the gene were constructed.Two different fragments were selected and inserted into pHANNIBAL flanking PDK intron in sense orientation and antisense orientation,thus forming hpRNA cassettes.Then the NotⅠfragment from pHANNIBAL containing the hpRNA cassettes were subcloned into the binary vector pART27.Finally the vectors were introduced into Agrobacterium LBA4404.These successfully constructed RNAi vectors and subsequent genetic transformation will provide useful clues for revealing the function of MS2-F gene during pollen development in flax.
