目的:探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)联合顺铂和IFN-γ应用对人骨肉瘤细胞生长抑制和诱导凋亡的作用。方法:用MTT测定TRAIL、顺铂、IFN-γ单独或联合应用对人骨肉瘤细胞MG-63的体外增殖抑制作用,并检测各组细胞凋亡率。结果:TRAIL、顺铂、IFN-γ三者联合作用于MG-63细胞后,能显著增强对MG-63的杀伤、抑制增殖及诱导凋亡作用,明显高于单独应用TRAIL组或顺铂组(P〈0.05)。结论:TRAIL、顺铂、IFN-γ三者联用能显著提高骨肉瘤细胞MG-63生长抑制和诱导凋亡作用。
Objective:To investigate the influence of tumor necrosis factor related apoptosis inducing ligand (TRAIL) combined with cisplatin (DDP) and IFN-γ on the proliferation and apoptosis of osteosarcoma cell MG-63. Methods: MTT assay was used to evaluate the cytotoxie effect on MG-63. Flow cytornetric analyses were performed to detect apoptosis rates. Results:The combined therapy induced significantly higher apoptosis and cytotoxicity than TRAIL, DDP or IFN-γ alone (P〈0.05). Conclusion:The combination of TRAIL, cisplatin and IFN-γ could significantly increase cytotoxic and apoptosis effect on MG-63.
