目的观察电离辐射对EL-4细胞survivin蛋白表达的影响及与P53表达的关系。方法采用流式细胞术(FCM)观察EL-4细胞接受4GyX射线照射后,survivin蛋白的表达、P53表达及细胞凋亡的变化。结果4Gy X射线照射EL-4细胞,Survivin蛋白表达受抑制,在4h明显降低,与对照组相比,差异有统计学意义(P〈0.001);照射组P53蛋白表达水平均高于对照组,在此时间点上与对照组相比,差异有统计学意义(P〈0.01)。照射后4h细胞凋亡率显著升高,与对照组相比差异有统计学意义(P〈0.001)。结论survivin与P53在细胞凋亡调控中发挥重要作用,survivin表达与P53表达水平相关。
Objective To explore the expression of survivin, an anti-apoptosis gene, in EL-4 cell line and its relationship with the expression of P53. Methods EL-4 cells were irradiated with 4 Gy X-rays to induce the expressions of survivin and P53. Flow cytometry assay was used to detect the expression of survivin and P53 and incidence of apoptosis following the irradiation. Results X-ray irradiation of 4 Gy down-regulated the expression of survivin in EL-4 cells while the expression of P53 was upregulated. A statistically lower expression of survivin was observed at 4 h after the irradiation when compared with the control group ( P 〈 0.001 ), while the expression of P53 was significantly increased in the exposed group ( P 〈 0.01 ). The incidence of apoptosis in the exposed group were significantly higher than that in the control group( P 〈 0.001 ). Conclusions It is shown that survivin and P53 play importam roles in regulating the cell apoptosis, and suggested that a possible correlation exists between expressions of survivin and P53.
